Usual Adult Dose for Chronic Idiopathic Constipation
2 mg orally once a day
Use: Treatment of chronic idiopathic constipation (CIC)
Renal Dose Adjustments
Mild to moderate renal dysfunction (CrCl at least 30 mL/min): No adjustment recommended.
Severe renal dysfunction (CrCl less than 30 mL/min): 1 mg orally once a day
End-stage renal disease requiring dialysis: Not recommended.

Precautions
CONTRAINDICATIONS:
- Hypersensitivity to the active component or any of the ingredients
- Intestinal perforation or obstruction due to structural/functional disorder of the gut wall, obstructive ileus, severe inflammatory conditions of the intestinal tract (e.g., Crohn's disease ulcerative colitis, toxic megacolon/megarectum)
Safety and efficacy have not been established in patients younger than pediatric patients.

Dialysis
End-stage renal disease requiring dialysis: Not recommended.

General:
- Doses exceeding 2 mg/day are not expected to increase efficacy.
- Safety and efficacy have been demonstrated in patients with chronic functional constipation; safety and efficacy have not been assessed in those with secondary causes of constipation (e.g., endocrine, metabolic, and/or neurologic disorders) or mediation-related constipation (e.g., opioid use). Some experts recommend that this drug should be avoided in patients with secondary causes of constipation.
Patient advice:
- Advise patients to speak to their healthcare provider if they become pregnant, intend to become pregnant, or are breastfeeding.
- Inform patients that this drug may cause dizziness and fatigue, and they should avoid driving or operating machinery if these side effects occur.
