Pirtobrutinib Side Effects

Applies to pirtobrutinib: oral tablet.

Serious side effects of Pirtobrutinib

Along with its needed effects, pirtobrutinib may cause some unwanted effects. Although not all of these side effects may occur, if they do occur they may need medical attention.

Check with your doctor immediately if any of the following side effects occur while taking pirtobrutinib:

More common

  • Black, tarry stools
  • bleeding gums
  • blood in the urine or stools
  • body aches or pain
  • chest pain or tightness
  • cough
  • ear congestion
  • fever or chills
  • loss of voice
  • pale skin
  • pinpoint red spots on the skin
  • sneezing
  • sore throat
  • stuffy or runny nose
  • swelling
  • trouble breathing
  • unusual bleeding or bruising
  • unusual tiredness or weakness

Less common

  • Confusion
  • dizziness
  • fainting
  • fast or irregular heartbeat
  • lightheadedness
  • lower back or side pain
  • painful or difficult urination
  • persistent non-healing sore
  • pink growth
  • reddish patch or irritated area
  • shiny bump
  • ulcers, sores, or white spots in the mouth
  • white, yellow, or waxy scar-like area

Other side effects of Pirtobrutinib

Some side effects of pirtobrutinib may occur that usually do not need medical attention. These side effects may go away during treatment as your body adjusts to the medicine. Also, your health care professional may be able to tell you about ways to prevent or reduce some of these side effects.

  • Burning, numbness, tingling, or painful sensations
  • constipation
  • cough
  • diarrhea
  • difficulty in moving
  • joint pain
  • large, flat, blue or purplish patches in the skin
  • muscle or bone pain
  • muscle stiffness
  • nausea
  • pain, swelling, or redness in the joints
  • stomach pain
  • unsteadiness or awkwardness
  • weakness in the arms, hands, legs, or feet

General

The most common adverse reactions reported were decreased neutrophils, hemoglobin, platelets, and lymphocytes, as well as fatigue, musculoskeletal pain, bruising, diarrhea, edema, dyspnea, and pneumonia.[Ref]

Dermatologic

Very common (10% or more): Rash (14%)[Ref]

Gastrointestinal

Very common (10% or more): Diarrhea (19%), constipation (13%), abdominal pain (11%), nausea (11%)[Ref]

Hematologic

Very common (10% or more): Hemorrhage (11%)[Ref]

Musculoskeletal

Very common (10% or more): Musculoskeletal pain (27%), arthritis/arthralgia (up to 12%)[Ref]

Nervous system

Very common (10% or more): Peripheral neuropathy (14%), dizziness (10%)

Ocular

Frequency not reported: Vision changes[Ref]

Oncologic

Frequency not reported: Tumor lysis syndrome[Ref]

Very common (10% or more): Fatigue (29%), edema (18%), fever (13%), pneumonia (16%), upper respiratory tract infections (10%), bruising (16%), decreased hemoglobin (42%), decreased platelet count (39%), decreased neutrophil count (36%), decreased lymphocyte count (32%), increased creatinine (30%), decreased calcium (19%), increased aspartate aminotransferase (AST) (17%), increased alanine transaminase (ALT) (11%), decreased potassium (13%), increased potassium (11%), decreased sodium (13%), increased lipase (12%), increased alkaline phosphatase (11%)

Common (1% to 10%): Sepsis (2.3%)
